We’d like to share new work from the University of Bergen detailing the development of a multiplexed imaging panel and bioinformatics pipeline, tested in head and neck squamous cell carcinoma, to characterize different tumor microenvironment components while maintaining a high degree of spatial detail. The approach, using Imaging Mass Cytometry™, holds promise for understanding the clinical implications of tumor heterogeneity in large patient cohorts using FFPE tissues available from biobanks worldwide. Here’s an application for identifying regions of immune activity and infiltration in breast tumor tissue, defining the highly heterogeneous nature of breast cancer. Samples were stained with a 41-marker panel comprised of the high-plex Human Immuno-Oncology IMC™ Panel, 31 Antibodies and expanded targets of interest, and acquired with whole slide imaging modes on Hyperion XTi™. Multiple types of lymphoid and myeloid immune cells are detected at the tumor margin (red inset). A tumor structure with low immune penetration is seen surrounding the necrotic tissue (cyan inset). Single-cell analysis quantitatively delineates cell phenotype of PD-L1-expressing tumor and immune cell populations.
